Spontaneous spike and burst activities in acute brain slices of adult mice

MEA is a rapidly expanding tool in electrophysiology that is used to study activity in neuronal circuits both in vitro and in vivo. It has been widely used to record different types of activity in cultured neurons and acute brain slices from young animals [1]. Acute brain slices from adult mice have been mostly used to study spontaneous activity during seizure-like conditions [2]. Under physiological conditions, spontaneous activity is very low in amplitude and is difficult to detect, particularly in acute brain slices from adult mice due to excessive cell death near the surface of the slice. Here we developed a protocol that largely overcomes this problem and thus allows the recording of spontaneous activity not only from young (P5 and P15) but also adult (P60-P75) acute brain slices. We took advantage of this protocol to investigate the developmental trajectory of the somatosensory network by analyzing the spike and burst rates and bursts synchronicity across different ages. In conclusion, we present a protocol that enables the recording of spontaneous activity under physiological conditions at different ages, which will provide insight into neuronal activity at the network level during development but also at later stages in models for neuropsychiatric and degenerative diseases.
